I. What Is Serum Separation Tube?

A serum separator tube is a container used to collect blood samples.

Contains a separator gel and a Clot activator.

The tube wall is siliconized and coated with a procoagulant to accelerate blood clotting and shorten the test time.

The tube is lined with a separating gel, which has a good affinity with the PET tube and does act as an isolator.

Generally, even on an ordinary centrifuge, the separating gel completely separates the liquid component (serum) from the solid component (blood cells) of the blood and accumulates in the tube to form a barrier.

No oil droplets are produced in the serum after centrifugation, and it is mainly used for serum biochemistry.

III. Reasons for poor separation of serum separation tubes

Serum separation gel procoagulant tubes are used to add a coagulant to blood collection tubes to shorten blood clotting time, obtain serum quickly, and report results in the shortest possible time.

Glass blood collection tubes do not need to add procoagulant; blood contact with the glass tube wall will trigger clotting.

In contrast, DEG and Ⅻ coagulation factors are very weakly activated when they come into contact with plastic blood collection tubes and require the addition of a procoagulant to shorten the clotting time.

In practice, it is often encountered that the separation of plastic blood collection tubes is not effective; for example: in some of the separation tubes can be seen in the separation of gel fragments or oil droplets floating on the surface of the serum or suspended in the serum; separation of gel layer floating in the serum layer above, etc...

The main reasons are as follows:

3.1 Quality of separation gel

The specific gravity of separation glue is between serum (plasma) and blood cells, which is the physical basis for the reversibility of separation glue and separation of serum (plasma).

Suppose the quality of the separation gel of the blood collection tube is poor.

In that case, the specific gravity does not meet the requirements and will inevitably affect the effect of the separation of serum (plasma); the phenomenon of separation gel and serum (plasma) intertwined is easy to appear.

3.2 Incomplete blood clotting

After centrifugation, sometimes there is incomplete separation of the gel compartment, serum, blood clot, and fibrin filaments appear in serum.

The reason is often that the blood is not completely coagulated before centrifugation. Incomplete blood coagulation can make fibrin mixed in the isolation layer.

It is necessary to use the serum separation tubes correctly according to the instructions and wait for the blood to be completely coagulated (generally, plastic tubes containing procoagulants need to be placed upright for about 30 min, and blood collection tubes without procoagulants need to be placed upright for 60-90 min) before centrifuging and separating the serum in order to prepare high-quality serum samples.

3.3 Centrifugation temperature

Centrifugation temperature significantly affects the effectiveness of separating serum from gel-promoting tubes.

Inert separator gel procoagulant tubes separated by normal centrifugation at room temperature showed clear serum, but small oily beads of varying sizes appeared in 15% to 20% of the samples.

In contrast, no oily beads were found in sera isolated from tubes centrifuged with a low-temperature centrifuge.

The inert separation gel will dissolve in the serum when the temperature exceeds the storage temperature required for the separation gel tubes.

It will not only block and contaminate the sample needle and reaction cup of the biochemical instrument but will also have a relatively large impact on the results of some biochemical measurements.

3.4 Centrifugation operation

If the centrifugal force is too low, the force acting on the separating gel is weak, making the separating gel poorly reversible, and it can appear that the fibrin clot stays in the serum or in the colloid layer, which can easily block the sample needle of the auto analyzer. On the contrary, if the centrifugation time is too long, the thixotropy is lost in the separation gel coagulum because the hydrogen bond is severed, and then it is difficult to form a mesh structure.
